- Method
- Flow cytometry using fluorescently labeled monoclonal antibodies against CD19, CD27, IgD, and IgM surface markers, followed by multiparameter analysis to enumerate switched-memory B cells (CD19+CD27+IgD-IgM+) per unit volume of whole blood.

- Specimen volume
- Minimum 3-5 mL whole blood for adults; 1-2 mL for pediatric patients
- Stability
- Specimen should be processed within 24-30 hours of collection when stored at room temperature (18-25°C). Do not refrigerate. If delay exceeds 24 hours, viability and surface antigen expression may be compromised.

Reference range
Reference values for CD19+CD27+IgD-IgM+ (class-switched memory B cells) in peripheral blood vary by laboratory and population. Published adult reference ranges generally fall between 10-80 cells/μL (absolute count) or 5-15% of total CD19+ B lymphocytes. Pediatric values differ and should be interpreted using age-appropriate reference data.N/A
No fasting is required. Patients should be well-hydrated. Recent vaccinations, immunosuppressive therapy, chemotherapy, or active infection may transiently alter B-cell subset distribution and should be documented at time of collection. Avoid collection within 24-48 hours of high-dose corticosteroid administration if possible.
