- Sterile sodium heparin (green-top) or EDTA (lavender-top) anticoagulated tube. ACD-A (yellow-top) tubes are also acceptable for flow cytometry immunophenotyping. Peripheral blood, bone marrow aspirate, or other body fluids may be submitted in sterile containers as appropriate for specimen type.

- Specimen volume
- Minimum 3-5 mL peripheral blood in adults; 1-3 mL in pediatric patients. Bone marrow aspirate: 1-2 mL in anticoagulant. Body fluids: aseptically collected volume per clinical indication. Specimens should be submitted in sufficient volume to allow for viability assessment and repeat analysis if needed.
- Stability
- Specimens should be processed within 24-48 hours of collection for optimal lymphocyte viability and antigen preservation. Ambient temperature transport (18-25°C) is preferred; do not refrigerate or freeze whole blood specimens intended for flow cytometry, as this may cause cell clumping, loss of viability, and altered antigen expression. If processing will be delayed beyond 48 hours, contact the performing laboratory for specific stability requirements.

Reference range
Reference ranges are laboratory-specific and vary based on population, age, and instrument platform. Results are typically reported as a percentage of CD25+CD19+ cells relative to total cells or lymphocytes analyzed. Consult the performing laboratory's reference interval for interpretation.N/A
No special patient preparation is required. Fasting is not necessary. Patients should inform their healthcare provider of any recent immunosuppressive therapy, corticosteroid use, chemotherapy, or biologic agent administration (e.g., rituximab, basiliximab, daclizumab), as these may alter lymphocyte subset distribution and affect test interpretation.
