What is this test?

The DOCK8-positive T cell test is a flow cytometry immune function assay. The test labels and analyzes immune cells in your blood, mainly T lymphocytes, to measure the expression of the DOCK8 protein within the CD3+CD14-CD45+ cell population. DOCK8 (Dedicator of Cytokinesis 8) is an important immune-regulating protein involved in signaling and functional regulation of immune cells.

Why might you need this test?

Your doctor may recommend this test to evaluate your immune system function. Abnormal DOCK8 protein expression may point to a primary immunodeficiency, such as DOCK8 deficiency (also known as autosomal dominant hyper-IgE syndrome). People with this condition often have recurrent infections, severe allergies, autoimmune disease, and an increased risk of certain types of tumors.

How is the test performed?

The test requires a venous blood sample. In the laboratory, the blood sample is incubated with specific fluorescently labeled antibodies and then analyzed with a flow cytometer. The instrument identifies and counts the percentage of T cells that express the DOCK8 protein. The process is similar to a routine blood draw and can usually be completed in an outpatient setting.

Reference range

Reference range: 95% of lymphocytes express DOCK8 (percent positive cells); Results are reported as percentage of CD3+CD14-CD45+ cells that are DOCK8+N/A

Specimen: whole-blood

No special preparation required. Standard venipuncture procedures apply. Avoid vigorous exercise immediately before sample collection.

Clinical interpretation

How abnormal results are interpreted and what conditions they suggest.

Frequently asked questions

Do I need to fast or make any special preparations before the test?
Generally, no special preparation is needed. You can eat and drink normally and take your regular medications as usual. If you have any questions, please check with your doctor before the test.
How long will it take to get my test results?
Flow cytometry testing usually takes 1-3 business days to complete. The exact turnaround time may vary depending on the schedule and workload of the laboratory performing the analysis.
Does an abnormal result mean I have a serious disease?
An abnormal result suggests a possible DOCK8-related immune deficiency, but a definite diagnosis requires your clinical symptoms, other laboratory tests, and genetic testing. If your result is abnormal, we recommend consulting an immunology or genetics specialist.
Can this test diagnose allergies?
DOCK8 testing is mainly used to evaluate immune deficiency rather than to diagnose allergies directly. However, people with DOCK8 deficiency often have severe allergic disease, so this test can be part of a comprehensive evaluation.
