Molecular target of the monoclonal antibody component

Belantamab mafodotin-blmf is indicated for the treatment of adult patients with relapsed or refractory multiple myeloma who have received at least four prior therapies, including an anti-CD38 monoclonal antibody, a proteasome inhibitor, and an immunomodulatory agent.

Belantamab mafodotin is an antibody-drug conjugate consisting of an afucosylated IgG1 antibody that specifically binds to BCMA (B-cell maturation antigen) expressed on multiple myeloma cells. Upon binding, the drug is internalized and releases the mcMMAF component, a microtubule inhibitor that disrupts cell division and causes cell death.

Common side effects include keratopathy (eye toxicity), thrombocytopenia, anemia, infusion-related reactions, and blurred vision. Regular ophthalmologic examinations are required during treatment. The drug may cause fetal harm and should not be used during pregnancy. It is administered intravenously by healthcare professionals.
