Treatment of Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML) in adult patients who are resistant or intolerant to prior therapy, including imatinib.

Bosutinib inhibits the BCR-ABL tyrosine kinase (the abnormal protein driving CML) and also blocks Src-family kinases including Src, Lyn, and Hck. This dual inhibition disrupts signals that promote cancer cell proliferation and survival.

Take with food; common side effects include diarrhea, nausea, and rash. Requires regular blood count and liver function monitoring. May cause fluid retention, QT prolongation, and cardiac events. Avoid in patients with hepatic impairment. Drug interactions with strong CYP3A inhibitors/inducers should be considered.
