Approved for the treatment of patients with advanced renal cell carcinoma (RCC), hepatocellular carcinoma (HCC) in patients previously treated with sorafenib, and progressive, metastatic medullary thyroid cancer (MTC).

Cabozantinib inhibits multiple receptor tyrosine kinases including MET, VEGFR-1/2/3, RET, KIT, TRKB, FLT-3, AXL, ROS1, TYRO3, MER, and TIE-2, disrupting tumor cell proliferation, survival, and angiogenesis (new blood vessel formation that feeds tumors).

Common side effects include diarrhea, fatigue, decreased appetite, hand-foot syndrome, and hypertension. Serious adverse events may include GI perforation, fistula formation, and hemorrhage. Requires regular monitoring of liver function and blood pressure. Should be taken on an empty stomach at least 2 hours before or 1 hour after eating.
