Calquence is FDA-approved for the treatment of adults with mantle cell lymphoma (MCL) who have received at least one prior therapy, as well as chronic lymphocytic leukemia (CLL) and small lymphocytic lymphoma (SLL). It may be used as monotherapy or in combination with other medications.
Calquence is a Bruton's tyrosine kinase (BTK) inhibitor. It works by irreversibly binding to and blocking the activity of BTK, an enzyme that plays a critical role in the survival, proliferation, and migration of abnormal B cells. By inhibiting this pathway, Calquence helps reduce the growth and spread of cancer cells.
Calquence may cause serious side effects including bleeding problems, infections, heart rhythm abnormalities (such as atrial fibrillation), cytopenias (low blood cell counts), and secondary malignancies. It should not be used during pregnancy as it may harm the fetus. Patients should inform their healthcare provider of all medications, as strong CYP3A inhibitors or inducers may interact with Calquence. Common side effects include headache, diarrhea, fatigue, and muscle pain.
