Capivasertib
Capivasertib is an AKT inhibitor used to treat certain types of breast cancer by targeting the PI3K/AKT signaling pathway. It works by blocking all three isoforms of the AKT protein kinase (AKT1, AKT2, AKT3), which are frequently overactive in tumors due to mutations or loss of tumor suppressors. This targeted therapy is typically used when cancer has progressed following prior endocrine therapy.
Capivasertib is an oral targeted therapy that inhibits AKT kinases for the treatment of hormone receptor-positive breast cancer with PI3K/AKT pathway alterations.

Capivasertib is indicated for the treatment of adult patients with hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative breast cancer that has progressed following treatment with an endocrine-based regimen.
Capivasertib is a potent inhibitor of all three isoforms of the serine/threonine kinase AKT (AKT1, AKT2, and AKT3). It blocks AKT signaling by preventing phosphorylation of downstream substrates, thereby inhibiting tumor cell growth and survival in cancers dependent on PI3K/AKT pathway activation.
Capivasertib may cause serious side effects including hyperglycemia, rash, and gastrointestinal toxicity. Regular monitoring of blood glucose and clinical response is recommended. Patients should be advised about potential drug interactions and the importance of compliance with dosing schedules.
