Treatment of leprosy (Hansen's disease), including lepromatous and multi-drug resistant leprosy, typically as part of combination therapy; sometimes used off-label for other mycobacterial infections such as disseminated MAC or drug-resistant tuberculosis.
Binds to mycobacterial DNA and inhibits bacterial RNA synthesis, suppressing growth of mycobacteria; also exhibits anti-inflammatory properties that help reduce immune-mediated tissue damage in leprosy.
Common side effects include skin discoloration (reddish-brown pigmentation), gastrointestinal symptoms, and photosensitivity; should be taken with food to improve absorption and reduce GI upset; contraindicated in patients with known hypersensitivity; may interact with other medications affecting QT interval.
