Published: July 26, 2026Updated: July 26, 2026

Crenessity (crinecerfont) is a corticotropin-releasing factor receptor 1 (CRF1) antagonist used to treat congenital adrenal hyperplasia (CAH). It works by blocking CRF signaling in the pituitary gland, which reduces the overproduction of adrenal androgens that are elevated in CAH. This medication is used in both adults and pediatric patients with classic and non-classic forms of CAH to help manage androgen excess.

Crenessity is indicated for the treatment of classic and non-classic congenital adrenal hyperplasia (CAH) in adults and pediatric patients 4 years and older. It is used to manage androgen excess in patients with CAH due to 21-hydroxylase deficiency.

Crenessity works as a corticotropin-releasing factor receptor 1 (CRF1) antagonist. By blocking CRF receptors in the pituitary gland, it reduces the excessive stimulation of ACTH secretion, which in turn decreases the overproduction of adrenal androgens.

Crenessity should be taken twice daily orally. Patients should be monitored for changes in adrenal hormone levels. Common side effects may include fatigue, headache, and dizziness. It is not a substitute for emergency glucocorticoid stress dosing in adrenal crisis situations.
