Danazol
Danazol is a synthetic androgen primarily used to treat endometriosis, fibrocystic breast disease, and hereditary angioedema. It works by suppressing gonadotropin secretion and creating a high-androgen, low-estrogen hormonal environment, which reduces estrogen-dependent tissue growth. This medication is typically reserved for patients who cannot tolerate other treatments or need specific therapeutic intervention for these conditions.
Danazol is an androgen derivative used to treat endometriosis, fibrocystic breast disease, and hereditary angioedema by altering hormonal balance.

Danazol is FDA-approved for treating moderate to severe endometriosis, fibrocystic breast disease (to reduce pain and nodularity), and hereditary angioedema (to prevent attacks). It may also be used off-label for other conditions involving abnormal estrogen activity.
Danazol suppresses pituitary gonadotropin secretion, leading to decreased estrogen production and increased androgen activity. This creates an unfavorable environment for estrogen-dependent tissues, reducing endometrial implants, breast nodules, and inflammatory episodes in hereditary angioedema.
Danazol carries significant androgenic side effects including acne, hirsutism, voice deepening, and weight gain. It is contraindicated during pregnancy (Category X), in patients with liver disease, and in those with androgen-dependent tumors. Regular monitoring of liver function and lipid panels is recommended during treatment.
