Danicopan is indicated as adjunctive therapy for the treatment of paroxysmal nocturnal hemoglobinuria (PNH) in adult patients with residual anemia despite treatment with a complement C5 inhibitor.
Danicopan reversibly binds to complement Factor D, preventing the cleavage of Factor B into Ba and Bb fragments, thereby selectively inhibiting the alternative complement pathway and reducing extravascular hemolysis.
Safety monitoring includes liver function tests and assessment for meningococcal infections. Not recommended as monotherapy for PNH. Use with caution in patients with active infections. Regular monitoring for hemolysis markers and transfusion requirements is advised.
