Danthron was used for short-term relief of constipation, including cases of opioid-induced constipation and for bowel preparation before diagnostic procedures or surgery. Its clinical use has been largely discontinued globally due to safety concerns.

As an anthraquinone derivative, Danthron stimulates peristalsis in the large intestine by irritating the bowel mucosa and increasing fluid and electrolyte secretion into the intestinal lumen, which accelerates stool passage.

Danthron has been withdrawn or severely restricted in many countries due to evidence of potential carcinogenicity (cancer risk) with prolonged use. It should only be used for short-term treatment under medical supervision and is contraindicated in patients with intestinal obstruction, inflammatory bowel disease, or abdominal pain of unknown origin.
