Denileukin Diftitox
Denileukin diftitox is a targeted fusion protein therapy that delivers the cytocidal portion of diphtheria toxin directly to cells expressing the IL-2 receptor. Primarily indicated for the treatment of cutaneous T-cell lymphoma (CTCL) in patients whose malignant T-cells express the CD25 component of the IL-2 receptor. May be considered in other hematologic malignancies expressing IL-2 receptors.
Denileukin diftitox is a fusion protein combining IL-2 with diphtheria toxin fragments. It binds specifically to cells expressing IL-2 receptors (CD25), undergoes internalization, and releases diphtheria toxin fragments that inhibit protein synthesis, leading to targeted cell death.
Administered via intravenous infusion by healthcare professionals experienced in chemotherapy. Common adverse effects include capillary leak syndrome, flu-like symptoms, and gastrointestinal disturbances. Pre-existing antibodies to diphtheria toxin may reduce efficacy. Requires careful monitoring of fluid status, kidney function, and vital signs during treatment.
