Fostamatinib inhibits spleen tyrosine kinase (SYK). Its active metabolite R406 blocks signal transduction from Fc-activating receptors and B-cell receptors, reducing immune-mediated destruction of platelets.
May cause liver injury, hypertension, diarrhea, and neutropenia. Requires regular monitoring of liver function and blood pressure. Drug interactions may occur with strong CYP3A4 inhibitors and inducers. Not recommended during pregnancy.
