Treatment of severe spasticity, particularly from multiple sclerosis, spinal cord injuries, amyotrophic lateral sclerosis, cerebral palsy, and other neurological conditions. Used when oral baclofen or other oral muscle relaxants are ineffective or cause intolerable side effects.
Gablofen is a GABA-B receptor agonist. When delivered intrathecally, it acts on spinal GABA-B receptors to inhibit the release of excitatory neurotransmitters, thereby reducing spasticity by decreasing the frequency and intensity of muscle contractions.
Must be administered by trained healthcare professionals through an implanted intrathecal pump. Abrupt discontinuation can cause severe withdrawal symptoms including seizures and organ failure. Dose adjustments require careful titration. Patients must maintain regular follow-up appointments for pump monitoring and refills. May cause drowsiness and interact with other CNS depressants.
