Halaven is FDA-approved for treating adults with metastatic breast cancer who have previously received at least two chemotherapy regimens (including an anthracycline and a taxane). It is also indicated for unresectable or metastatic liposarcoma in patients who have received prior chemotherapy.
Halaven is a microtubule inhibitor and a synthetic analog of halichondrin B. It works by binding to tubulin and inhibiting microtubule polymerization, which prevents the formation of the mitotic spindle needed for cancer cell division. This ultimately leads to cell cycle arrest and apoptosis in cancer cells.
Halaven is administered by intravenous infusion, typically on specific days of a treatment cycle. Common side effects include neutropenia, fatigue, nausea, hair loss, and peripheral neuropathy. It may cause serious side effects including severe neutropenia and QT prolongation. Patients should be monitored closely during treatment, and dosage adjustments may be needed based on blood counts and other factors.
