Published: July 26, 2026Updated: August 25, 2026

Infigratinib is an oral targeted therapy known as an FGFR (fibroblast growth factor receptor) inhibitor used to treat certain cancers with specific genetic alterations. It works by blocking FGFR enzymes that drive tumor growth in cancers with FGFR alterations. It is primarily used in patients with advanced or metastatic cholangiocarcinoma who have FGFR2 gene fusions or rearrangements and have received prior treatment.

Quick summary

FDA-approved for treatment of adults with previously treated, unresectable locally advanced or metastatic cholangiocarcinoma harboring FGFR2 fusions or rearrangements. May also be used in clinical trials or off-label for other advanced solid tumors with FGFR genetic alterations.

Infigratinib is a small molecule tyrosine kinase inhibitor that selectively blocks FGFR1, FGFR2, and FGFR3 enzymes. By inhibiting these receptors, it interrupts signaling pathways that promote cancer cell growth, survival, and proliferation in tumors with FGFR alterations.

Common side effects include hyperphosphatemia, nail changes, stomatitis, and fatigue. Requires monitoring of phosphate levels and liver function. May cause ocular toxicity including retinal pigment epithelial detachment. Contraindicated in pregnancy due to fetal harm. Patients should be assessed for FGFR alterations before treatment initiation.
