Inrebic is indicated for the treatment of adult patients with intermediate-2 or high-risk primary myelofibrosis (also known as chronic idiopathic myelofibrosis) and for the treatment of adult patients with intermediate-2 or high-risk secondary myelofibrosis (post-polycythemia vera and post-essential thrombocythemia).

Inrebic is a Janus kinase 2 (JAK2) inhibitor. It works by blocking the activity of JAK2, a protein that is often mutated and overactive in myelofibrosis. By inhibiting this overactive signaling pathway, the medication helps reduce the abnormal blood cell production and symptoms associated with the disease, such as an enlarged spleen and constitutional symptoms.

Inrebic carries a boxed warning for serious and fatal cases of Wernicke's encephalopathy, a neurological emergency caused by thiamine deficiency. Patients should have thiamine levels monitored before and during treatment. Other common side effects include diarrhea, nausea, vomiting, and anemia. The medication may cause serious liver problems and should be used with caution in patients with renal or hepatic impairment. Inrebic is typically taken once daily with or without food and should not be used in patients with severe renal impairment.
