Used for the treatment of adult and pediatric patients with solid tumors that have an NTRK gene fusion. This includes various cancer types such as lung cancer, thyroid cancer, salivary gland cancer, sarcoma, and other solid tumors, regardless of their location in the body.
Larotrectinib is a selective inhibitor of tropomyosin receptor kinases (TRKA, TRKB, TRKC). It binds to these receptors with high affinity (IC50 values of 5-11 nM), blocking their signaling pathways that promote cancer cell growth and survival.
Common side effects may include fatigue, dizziness, nausea, and neurological effects. It is an oral medication taken twice daily. Patients should have their tumor tested for NTRK gene fusion before starting treatment. May cause serious adverse effects including neurotoxicity and hepatotoxicity in some patients.
