Lomustine
Lomustine is an oral alkylating agent classified as a nitrosourea used in the treatment of certain cancers. It works by damaging DNA and RNA in cancer cells through alkylation, preventing their growth and division. It is primarily used in patients with brain tumors, Hodgkin's lymphoma, and other malignancies.
Lomustine is an oral chemotherapy drug that alkylates DNA to treat certain brain tumors and lymphomas, requiring careful monitoring due to significant side effects.

Lomustine is indicated for the treatment of brain tumors (including glioblastoma multiforme and astrocytoma), Hodgkin's lymphoma (typically in combination with other chemotherapeutic agents), melanoma, and non-small cell lung cancer.
As a nitrosourea alkylating agent, lomustine transfers alkyl groups to DNA and RNA molecules, causing cross-linking of DNA strands that prevents cancer cell replication. It may also inhibit key enzymatic processes through carbamoylation of proteins.
Lomustine can cause significant bone marrow suppression with delayed nadir (low blood counts occurring 4-6 weeks after dosing), requiring regular blood monitoring. Other important adverse effects include pulmonary fibrosis, hepatotoxicity, and nephrotoxicity. Treatment is typically given in cycles with extended monitoring due to cumulative and delayed toxicities.
