Loncastuximab Tesirine
Loncastuximab tesirine is a targeted antibody-drug conjugate (ADC) used to treat adults with relapsed or refractory large B-cell lymphoma, including diffuse large B-cell lymphoma (DLBCL), after at least two prior lines of systemic therapy. It works by binding to CD19, a protein found on the surface of B-cells, and delivering a cytotoxic agent directly to cancer cells to destroy them. This medication provides an important treatment option for patients whose cancer has returned or not responded to previous therapies.

Treatment of adults with relapsed or refractory large B-cell lymphoma (including diffuse large B-cell lymphoma, DLBCL) after two or more lines of systemic therapy.
An antibody-drug conjugate (ADC) that binds to CD19 antigen on the surface of B-cells. Once bound, it internalizes and releases the cytotoxic payload SG3199 (a PBD dimer), which causes DNA crosslinking and triggers apoptosis (programmed cell death) of malignant B-cells.
Administered as an intravenous infusion every 3 weeks. Associated with boxed warnings for fluid retention and effusion, and liver toxicity. Pre-medication with dexamethasone is required. Common side effects include thrombocytopenia, neutropenia, anemia, and elevated gamma-glutamyl transferase. Requires dose adjustments for certain adverse reactions.
