Treatment of adults with ALK-positive metastatic non-small cell lung cancer (NSCLC) who have progressed on or are intolerant to previous ALK inhibitor therapy such as crizotinib, alectinib, or brigatinib.

Lorlatinib inhibits anaplastic lymphoma kinase (ALK) and ROS1 receptors, blocking signaling pathways that cancer cells use to grow, divide, and spread. It also targets other kinases including TRK and FAK, and remains active against several ALK resistance mutations.

Common side effects include edema, weight gain, neuropathy, and cognitive effects. Requires regular monitoring of cholesterol, triglycerides, and liver function. May cause serious CNS side effects and should be used with caution in patients with cardiovascular disease. Must be taken consistently with or without food.
