Treatment of schizophrenia in adults; treatment of depressive episodes associated with bipolar I or II disorder as monotherapy or as adjunctive therapy with lithium or valproate; and as adjunctive therapy with antidepressants.

Lumateperone is a dopamine D2 receptor antagonist and serotonin 5-HT2A receptor antagonist. It also exhibits activity at 5-HT1A receptors, D1 receptors, and acts as a serotonin reuptake inhibitor, providing a multimodal mechanism that differentiates it from other antipsychotics.

Common side effects may include somnolence, dry mouth, and dizziness. It is not approved for use in elderly patients with dementia-related psychosis due to increased mortality risk. Patients should be monitored for metabolic changes, weight gain, and movement disorders. Avoid use during pregnancy unless benefits outweigh risks.
