Moxetumomab Pasudotox
Moxetumomab pasudotox is a CD22-directed immunotoxin used to treat relapsed or refractory hairy cell leukemia in adult patients who have received at least two prior lines of therapy. It works by targeting CD22 on cancer cells and delivering a bacterial toxin that inhibits protein synthesis, leading to cell death. This medication is administered intravenously and requires careful monitoring for capillary leak syndrome and hemolytic uremic syndrome.

Treatment of adult patients with relapsed or refractory hairy cell leukemia who have received at least two prior systemic therapies, including purine nucleoside analogs.
Moxetumomab pasudotox is a recombinant immunotoxin consisting of an anti-CD22 antibody fused to a truncated Pseudomonas exotoxin. It binds specifically to CD22 on B-cell surfaces, undergoes internalization, and releases the toxin component which ADP-ribosylates elongation factor 2, inhibiting protein synthesis and inducing apoptosis of malignant B cells.
Requires monitoring for capillary leak syndrome and hemolytic uremic syndrome. Contraindicated in patients with known hypersensitivity. Common adverse effects include peripheral edema, nausea, fatigue, and pyrexia. Should be administered in healthcare settings with appropriate resuscitation equipment available. Female patients of reproductive potential should use effective contraception during and for a period after treatment.
