Nintedanib is approved for treating idiopathic pulmonary fibrosis (IPF), other progressive fibrosing interstitial lung diseases (PF-ILDs), and systemic sclerosis-associated interstitial lung disease (SSc-ILD). It is also used in combination with other medications for certain types of lung cancer.

Nintedanib inhibits multiple receptor tyrosine kinases (RTKs) including platelet-derived growth factor receptors (PDGFR α/β), fibroblast growth factor receptors (FGFR 1-3), and vascular endothelial growth factor receptors (VEGFR). By blocking these pathways involved in fibroblast proliferation, angiogenesis, and tissue remodeling, it reduces fibrotic processes in the lungs.

Common side effects include diarrhea, nausea, abdominal pain, liver enzyme elevations, and vomiting. Regular liver function monitoring is required during treatment. Nintedanib may cause fetal harm and is contraindicated during pregnancy. Patients with moderate to severe hepatic impairment should use with caution.
