Oliceridine

Published: July 26, 2026Updated: July 26, 2026

Oliceridine is a full opioid agonist that selectively targets the mu-opioid receptor to provide potent pain relief for acute pain severe enough to require an opioid analgesic. Unlike partial agonists, it has no ceiling effect, meaning that higher doses can provide greater analgesia within safe limits. This medication is administered intravenously in clinical settings.

Quick summary

Oliceridine is an IV opioid analgesic for acute pain that works as a full mu-opioid receptor agonist with no ceiling effect.

Overdose information

Important overdose information
Toxic dose: Doses significantly exceeding therapeutic limits (typically above 5mg in opioid-naive individuals) may cause serious respiratory depression and CNS depression
Chronic overdose: Prolonged exposure to high cumulative doses may lead to tolerance, dependence, and potential organ damage from hypoxia
Immediate actions
Ensure airway and breathing support, administer naloxone, monitor vital signs, consider gastric decontamination if ingestion was recent, provide oxygen therapy
Antidote
Naloxone(Highly effective in reversing opioid effects; may require repeated dosing due to shorter half-life compared to oliceridine)
Poison Control: 1-800-222-1222

Delivery method

Management of acute pain severe enough to require an opioid analgesic in adult patients, typically in hospital or clinical settings.

Oliceridine is a full agonist at the mu-opioid receptor. By binding to these receptors in the brain and spinal cord, it activates pain-inhibiting pathways and reduces the perception of pain signals. As a full agonist, it provides maximum analgesic effect without the ceiling limitation seen with partial agonists.

Administered via intravenous route only. Like all opioids, it carries risks of dependence, respiratory depression, and other opioid-related adverse effects. Dosage should be individually titrated based on patient response and pain severity.
