Paltusotine
Paltusotine is a selective somatostatin receptor 2 (SSTR2) agonist that suppresses growth hormone (GH) and insulin-like growth factor-1 (IGF-1) secretion. It is being investigated for the treatment of acromegaly and neuroendocrine tumors that involve excess hormone secretion. The medication works by mimicking the natural hormone somatostatin, providing an alternative to injectable somatostatin analogues.
A targeted therapy that suppresses growth hormone and IGF-1 by selectively activating somatostatin receptor 2, primarily under investigation for acromegaly and neuroendocrine tumors.

Under investigation for acromegaly (a condition characterized by excess growth hormone in adults) and potentially for neuroendocrine tumors that overproduce hormones. It may offer an oral alternative to traditional injectable somatostatin analogues.
Paltusotine selectively binds to and activates somatostatin receptor 2 (SSTR2) with over 4000-fold greater affinity for this receptor compared to other somatostatin receptor subtypes. This activation inhibits GH release from the pituitary gland, thereby reducing IGF-1 levels produced by the liver.
As an investigational drug, paltusotine is not yet FDA-approved. Side effects may be similar to other somatostatin analogues and could include gastrointestinal symptoms, gallbladder abnormalities, and blood glucose changes. Patients should use only under physician supervision and according to prescribed treatment protocols.
