Panobinostat
Panobinostat is a histone deacetylase (HDAC) inhibitor used in the treatment of multiple myeloma, particularly in patients who have received at least two prior therapies including bortezomib. Treatment of relapsed and/or refractory multiple myeloma in adult patients who have received at least two prior regimens including bortezomib and an immunomodulatory agent. Typically used in combination with bortezomib and dexamethasone.
Panobinostat is a histone deacetylase (HDAC) inhibitor that blocks HDAC enzymes, leading to increased histone acetylation. This alters gene expression patterns, induces cell cycle arrest, and promotes apoptosis in cancer cells, particularly myeloma cells.
Administered orally. Common side effects include thrombocytopenia, diarrhea, fatigue, and nausea. Requires monitoring of blood counts and electrolytes. Contraindicated in patients with certain heart conditions. Drug interactions possible with strong CYP3A4 inhibitors and inducers.
