Pemigatinib
Pemigatinib is a targeted therapy medication that inhibits fibroblast growth factor receptors (FGFR1, 2, and 3), blocking signals that promote cancer cell growth and survival. It is primarily used to treat adults with advanced cholangiocarcinoma (bile duct cancer) who have FGFR2 gene alterations and have received prior systemic therapy.
Pemigatinib is an FGFR inhibitor used to treat FGFR-altered cholangiocarcinoma and other FGFR-driven cancers.

Pemigatinib is indicated for the treatment of adults with previously treated, unresectable locally advanced or metastatic cholangiocarcinoma with FGFR2 fusion or other rearrangement. It may also be used for other malignancies harboring FGFR genetic alterations.
Pemigatinib is a small molecule kinase inhibitor that selectively blocks FGFR1, 2, and 3 by inhibiting receptor phosphorylation. This disrupts downstream signaling pathways involved in tumor cell proliferation, survival, and angiogenesis in cancers with FGFR alterations.
Common adverse reactions include hyperphosphatemia, nail disorders, alopecia, and visual disturbances such as retinal pigment epithelial detachment. Requires monitoring of phosphate levels and ophthalmologic exams. Contraindicated in pregnancy due to risk of fetal harm. Should be avoided in patients with severe hepatic or renal impairment.
