Published: July 26, 2026Updated: August 20, 2026

Penbutolol is a non-selective beta-blocker medication used primarily to treat high blood pressure (hypertension). It works by blocking the effects of adrenaline on beta-adrenergic receptors in the heart and blood vessels, which reduces heart rate and blood pressure. This medication may be prescribed when other first-line antihypertensives are not adequately controlling blood pressure or are not well-tolerated.

Penbutolol is primarily indicated for the treatment of essential hypertension (high blood pressure). It may be used as monotherapy or in combination with other antihypertensive agents when single-agent therapy is insufficient.

Penbutolol is a non-selective beta-blocker that competitively antagonizes both beta-1 and beta-2 adrenergic receptors. It reduces heart rate, decreases cardiac output, and lowers blood pressure. The medication also possesses intrinsic sympathomimetic activity (ISA), meaning it has partial agonist properties at beta receptors.

Penbutolol should not be discontinued abruptly as this may precipitate angina, myocardial infarction, or rebound hypertension. It should be used with caution in patients with asthma, diabetes, or heart failure. Common side effects may include fatigue, bradycardia, and cold extremities. This medication is contraindicated in patients with severe bradycardia, AV block, or decompensated heart failure.
