Treatment of symptomatic tenosynovial giant cell tumor (TGCT), also known as pigmented villonodular synovitis (PVNS), in adults where surgery would cause functional limitation or severe morbidity.

Pexidartinib inhibits colony stimulating factor 1 receptor (CSF1R), KIT, and FLT3 with ITD mutations. By blocking CSF1R, it reduces the abnormal cell proliferation that drives TGCT tumor growth.

Associated with risk of serious and potentially fatal liver injury requiring regular hepatic monitoring. Contraindicated in patients with pre-existing severe hepatic impairment. May cause fetal harm; contraception required during treatment and for one month after. Available only through a restricted distribution program.
