Treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL) in adults, typically given in combination with bendamustine and rituximab for patients who have received at least two prior therapies.
Polatuzumab vedotin is an antibody-drug conjugate that binds to CD79b, a protein found on the surface of B cells. Once bound, the conjugate is internalized and releases the cytotoxic payload MMAE (monomethyl auristatin E), which inhibits cell division and triggers apoptosis (programmed cell death) in malignant B cells.
Common side effects include peripheral neuropathy, neutropenia, thrombocytopenia, anemia, fatigue, and infusion-related reactions. Patients should be monitored for liver toxicity and infections. Use with caution in patients with pre-existing neuropathy. Pregnancy should be avoided during treatment due to potential harm to the fetus.
