Treatment of chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) in adult patients who have failed prior tyrosine kinase inhibitor therapy, including those with the T315I mutation.
Ponatinib inhibits the activity of the ABL tyrosine kinase, including the T315I mutant form, as well as additional kinases involved in cancer cell proliferation and survival. By blocking these signaling pathways, it reduces the growth and spread of leukemia cells.
Requires prescription and supervision by an oncologist. Has significant cardiovascular risks including arterial occlusion and heart failure. Regular monitoring of cardiac function, blood pressure, and liver function is necessary during treatment.
