Scemblix is indicated for the treatment of adult patients with Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML) in chronic phase who have been previously treated with two or more tyrosine kinase inhibitors. It is also approved for patients with the T315I mutation.

Scemblix is a tyrosine kinase inhibitor that works by specifically binding to the ABL myristoyl pocket (STAMP mechanism), which differs from traditional TKIs that bind to the ATP site. This allows it to inhibit BCR-ABL1 activity even in cases where other tyrosine kinase inhibitors are no longer effective.

Scemblix is taken orally, typically twice daily. Common side effects may include musculoskeletal pain, fatigue, and low blood cell counts. Patients should be monitored for heart function and blood pressure changes. It may interact with other medications and should be taken as prescribed by a healthcare provider.
