Sebetralstat

Published: July 26, 2026Updated: July 26, 2026

Sebetralstat is an oral on-demand treatment for hereditary angioedema (HAE) that works by inhibiting plasma kallikrein, the enzyme responsible for producing bradykinin. By blocking this pathway, it reduces the bradykinin-mediated swelling attacks characteristic of HAE.

Quick summary

Sebetralstat is an oral plasma kallikrein inhibitor used for on-demand treatment of hereditary angioedema attacks.

Sebetralstat is indicated for the on-demand treatment of hereditary angioedema (HAE) attacks in adults and children. HAE is a rare genetic disorder caused by C1 inhibitor deficiency, leading to recurrent episodes of swelling in various parts of the body including the abdomen, face, hands, feet, and throat.

Sebetralstat is a competitive, reversible inhibitor of plasma kallikrein, a serine protease. It blocks the enzyme from cleaving high molecular weight kininogen (HK), which prevents the release of bradykinin. Since bradykinin is the key mediator that increases vascular permeability and causes swelling in HAE, inhibiting its production helps reduce attack severity and duration.

Sebetralstat is taken orally at the onset of an HAE attack, making it a convenient option for on-demand treatment. It is not intended for prophylaxis (routine prevention). Patients should have an updated HAE action plan and seek emergency care for throat swelling or severe abdominal attacks. As with all HAE treatments, individual response may vary, and this medication should be used under healthcare provider guidance.
