Seglentis is indicated for the management of acute pain severe enough to require an opioid analgesic and for which alternative treatments are inadequate. It is approved for use in adults and is typically prescribed for short-term treatment (typically 14 days or less).

Seglentis combines two active ingredients with complementary mechanisms: celecoxib, a COX-2 selective nonsteroidal anti-inflammatory drug (NSAID) that reduces pain and inflammation by inhibiting the cyclooxygenase-2 enzyme; and tramadol, an atypical opioid analgesic that binds to mu-opioid receptors and inhibits reuptake of norepinephrine and serotonin to modulate pain signals.

Seglentis carries a boxed warning for addiction, abuse, and misuse risks, as well as serious respiratory depression and opioid use disorder risks. It should not be used in patients with known GI bleeding, severe renal impairment, or in the perioperative period of coronary artery bypass graft surgery. Common side effects include nausea, headache, dizziness, and somnolence. Seglentis is classified as a Schedule IV controlled substance due to the tramadol component. Use may increase risk of serotonin syndrome and seizures.
