Sotorasib
Sotorasib is a targeted cancer therapy that inhibits the KRAS G12C mutant protein, a driver of tumor growth in certain cancers. It is approved for use in adults with advanced non-small cell lung cancer (NSCLC) that has a KRAS G12C mutation and has progressed after at least one prior systemic therapy.
Sotorasib is an oral KRAS G12C inhibitor used to treat mutant KRAS-positive non-small cell lung cancer.

Treatment of adults with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) who have received at least one prior systemic therapy. Requires confirmation of KRAS G12C mutation through FDA-approved testing before use.
Sotorasib is a KRAS G12C inhibitor that forms an irreversible, covalent bond with the unique cysteine residue of the mutant KRAS G12C protein, locking it in an inactive GDP-bound state and preventing downstream oncogenic signaling pathways that drive tumor growth.
Taken orally once daily with or without food. Common side effects include diarrhea, fatigue, nausea, and musculoskeletal pain. May cause hepatotoxicity requiring monitoring of liver function. Patients should be advised to report any signs of interstitial lung disease. Effectiveness requires KRAS G12C mutation confirmation prior to treatment initiation.
