Sporanox treats systemic fungal infections such as aspergillosis, blastomycosis, histoplasmosis, and paracoccidioidomycosis. It is also used for oropharyngeal and esophageal candidiasis, as well as dermatophyte infections of the nails (onychomycosis).
Itraconazole works by inhibiting the enzyme lanosterol 14α-demethylase, which is essential for producing ergosterol—a critical component of fungal cell membranes. This disruption weakens the cell membrane, preventing fungal growth and replication.
Sporanox has significant drug interactions and is contraindicated with certain medications including simvastatin, lovastatin, quinidine, and ergot alkaloids. It may worsen heart failure and should not be used in patients with known ventricular dysfunction. Liver function should be monitored during treatment. The oral capsules and oral solution are not interchangeable.
