Tazemetostat
Tazverik (tazemetostat) is a targeted therapy medication that inhibits the EZH2 enzyme, which is involved in gene expression regulation. It is prescribed for adults and teenagers (16 years and older) with metastatic or locally advanced epithelioid sarcoma who cannot have complete surgical removal, and for adults with relapsed or refractory follicular lymphoma with specific EZH2 mutations or who have exhausted other treatment options.
Tazverik is an EZH2 inhibitor used to treat certain types of epithelioid sarcoma and follicular lymphoma by targeting cancer cell growth.

Tazverik is indicated for the treatment of adults and pediatric patients (16 years and older) with metastatic or locally advanced epithelioid sarcoma not eligible for complete surgical resection, as well as for adults with relapsed or refractory follicular lymphoma whose tumors harbor an EZH2 mutation (after at least two prior systemic therapies) or those who have no satisfactory alternative treatment options.
Tazverik works as a selective inhibitor of the EZH2 enzyme, which is a histone methyltransferase. By blocking EZH2 activity, the medication reduces methylation of histone H3 lysine 27 (H3K27), leading to altered gene expression and decreased proliferation of cancer cells that depend on this pathway.
Tazverik is taken orally, typically twice daily with or without food. Common side effects include fatigue, nausea, abdominal pain, and musculoskeletal pain. The medication may increase the risk of secondary malignancies due to its mechanism of action. Regular monitoring and blood tests may be required during treatment. Patients should discuss all medications and supplements with their healthcare provider to avoid interactions.
