Tegsedi is indicated for the treatment of stage 1 or stage 2 polyneuropathy in adults with hereditary transthyretin amyloidosis (hATTR). This rare genetic disorder causes abnormal amyloid protein to accumulate in various organs, particularly affecting the peripheral nervous system.

Tegsedi contains the active ingredient inotersen, an antisense oligonucleotide that specifically binds to messenger RNA (mRNA) produced by the transthyretin (TTR) gene. By targeting and degrading this mRNA, the medication reduces the production of the TTR protein, thereby limiting the formation of amyloid deposits that cause organ and nerve damage.

Tegsedi requires regular monitoring, including weekly blood tests for platelet counts and kidney function. Common side effects may include injection site reactions, nausea, and fatigue. The medication carries a boxed warning for serious risks of thrombocytopenia and glomerulonephritis, which require prompt medical attention. Patients should be monitored for vitamin A levels and liver function during treatment.
