Trazimera is indicated for the treatment of HER2-positive breast cancer (early-stage, metastatic, and as adjuvant therapy) and HER2-positive metastatic gastric or gastroesophageal junction adenocarcinoma. Treatment is typically reserved for patients whose tumors demonstrate HER2 protein overexpression or HER2 gene amplification.
Trazimera is a monoclonal antibody that binds specifically to the HER2 (Human Epidermal Growth Factor Receptor 2) protein on the surface of cancer cells. By binding to HER2, it inhibits the signaling pathways that promote cancer cell proliferation and survival. Additionally, it may trigger antibody-dependent cell-mediated cytotoxicity (ADCC) to help destroy cancer cells.
Trazimera carries a boxed warning for cardiotoxicity, including decreased left ventricular ejection fraction and heart failure; cardiac monitoring is required during treatment. It may cause serious infusion reactions, and patients should be monitored during and after infusions. As a biosimilar, Trazimera is approved based on demonstrated similarity to the reference product Herceptin in terms of efficacy, safety, and purity.
