Pexidartinib
Turalio (pexidartinib) is a prescription tyrosine kinase inhibitor used to treat adults with symptomatic tenosynovial giant cell tumor (TGCT), a rare non-cancerous tumor affecting the joints and tendon sheaths. By blocking the CSF1 receptor, it helps reduce tumor growth and associated symptoms like pain, swelling, and limited movement.
Turalio is a targeted therapy for tenosynovial giant cell tumor that works by blocking the CSF1 signaling pathway to reduce tumor size and symptoms.

Turalio is indicated for the treatment of symptomatic tenosynovial giant cell tumor (TGCT) in adults with severe morbidity or functional limitations who are not candidates for surgery. TGCT, also known as pigmented villonodular synovitis (PVNS), causes abnormal tissue growth in joints and tendon sheaths.
Turalio works by inhibiting the colony-stimulating factor 1 receptor (CSF1R), a tyrosine kinase receptor that plays a key role in the growth and survival of cells that drive TGCT. By blocking this signaling pathway, the medication helps reduce the proliferation of tumor cells and associated inflammation.
Turalio carries a boxed warning for hepatotoxicity (liver damage), and liver function must be monitored regularly during treatment. Common side effects may include changes in hair color, fatigue, rash, and digestive issues. It is only available through a restricted distribution program due to its serious risks. Patients should discuss all potential side effects and necessary monitoring with their healthcare provider.
