Umbralisib is indicated for the treatment of adult patients with relapsed or refractory marginal zone lymphoma (MZL) and relapsed or refractory follicular lymphoma (FL) who have received at least one prior anti-CD20-based regimen.

Umbralisib is a phosphoinositide 3-kinase (PI3K) delta inhibitor. It works by blocking the PI3K-delta pathway, which is involved in the proliferation, survival, and migration of malignant B-cells. By inhibiting this signaling pathway, the drug helps control the growth and spread of cancer cells.

Common side effects include diarrhea, fatigue, nausea, and liver enzyme elevations. It carries a FDA Boxed Warning for hepatotoxicity and serious diarrhea/colitis. Patients require regular monitoring of liver function tests. May increase risk of infections and should be used with caution in patients with underlying immune system disorders. Drug interactions may occur with strong CYP3A4 inducers and inhibitors.
