Velcade is indicated for the treatment of multiple myeloma (including newly diagnosed and relapsed/refractory cases) and mantle cell lymphoma. It may be used as monotherapy or in combination with other cancer medications.
Velcade is a proteasome inhibitor that reversibly blocks the 26S proteasome, a cellular complex responsible for degrading proteins. This inhibition disrupts multiple signaling pathways within cancer cells, leading to accumulation of misfolded proteins, cell cycle arrest, and programmed cell death (apoptosis).
Velcade is administered via subcutaneous or intravenous injection by healthcare professionals. Common side effects include nausea, fatigue, peripheral neuropathy, and low blood counts. It may cause low blood pressure and cardiac effects. Patients should be monitored regularly for hematologic toxicity and liver function. Pregnancy category D - should not be used during pregnancy.
