Larotrectinib
Vitrakvi (larotrectinib) is a targeted cancer therapy known as a TRK inhibitor. It is used to treat adults and children with solid tumors that have a neurotrophic receptor tyrosine kinase (NTRK) gene fusion—a genetic abnormality where NTRK genes fuse with other genes, producing proteins that drive cancer growth. It is typically used when the cancer has spread or cannot be surgically removed and there are no satisfactory alternative treatments.
Vitrakvi is a targeted therapy for cancers with NTRK gene fusions, approved for both adult and pediatric patients with solid tumors.

Vitrakvi is indicated for the treatment of adult and pediatric patients with solid tumors that have a neurotrophic receptor tyrosine kinase (NTRK) gene fusion without a known acquired resistance mutation, are metastatic or where surgical resection is likely to result in severe morbidity, and have no satisfactory alternative treatments or whose cancer has progressed following treatment.
Vitrakvi is a tropomyosin receptor kinase (TRK) inhibitor. It works by blocking the activity of TRK proteins (TRKA, TRKB, TRKC) produced by NTRK genes. When NTRK genes are fused with other genes, they produce abnormal TRK proteins that promote uncontrolled cancer cell growth. By inhibiting these TRK proteins, Vitrakvi can slow or stop tumor growth.
Vitrakvi is available as both oral capsules and liquid formulation for pediatric patients. It is a prescription medication that requires genetic testing to confirm NTRK gene fusion before use. Common side effects may include fatigue, dizziness, nausea, and vomiting. Patients should discuss potential side effects and drug interactions with their healthcare provider.
