Vosevi is indicated for the treatment of chronic hepatitis C virus (HCV) infection in adults with genotypes 1-6. It is specifically approved for patients who have previously been treated with a sofosbuvir-containing regimen without an NS5A inhibitor, and may be used in other appropriate cases as determined by a healthcare provider.
Vosevi combines three direct-acting antiviral agents: sofosbuvir (a nucleotide analog inhibitor targeting NS5B polymerase), velpatasvir (an NS5A inhibitor), and voxilaprevir (a protease inhibitor targeting NS3/4A). Together, these components interfere with multiple stages of the HCV replication cycle, effectively suppressing viral load.
Vosevi is contraindicated in patients with moderate to severe hepatic impairment (Child-Pugh B and C) due to risk of serious liver problems. It should not be used during pregnancy or in combination with certain medications that significantly reduce its effectiveness. Common side effects include headache, fatigue, nausea, diarrhea, and insomnia. Patients should be monitored for potential reactivation of hepatitis B in those with prior HBV infection.
