Belzutifan
Welireg (belzutifan) is an oral medication that inhibits hypoxia-inducible factor 2 alpha (HIF-2α), a protein that promotes tumor growth. It is prescribed for adults with Von Hippel-Lindau disease who require treatment for associated renal cell carcinoma, central nervous system hemangioblastomas, or pancreatic neuroendocrine tumors, and for adults with advanced renal cell carcinoma following prior therapy.
Welireg is an oral HIF-2α inhibitor used to treat Von Hippel-Lindau disease-related tumors and advanced kidney cancer.

Treatment of adults with Von Hippel-Lindau (VHL) disease who require therapy for associated renal cell carcinoma, CNS hemangioblastomas, or pancreatic neuroendocrine tumors; and for adults with advanced renal cell carcinoma after prior anti-angiogenic therapy.
Welireg blocks hypoxia-inducible factor 2 alpha (HIF-2α), a transcription factor that drives tumor growth by regulating genes involved in cell proliferation, angiogenesis, and metabolism. In VHL disease, defective VHL protein leads to abnormal HIF-2α accumulation, and inhibiting this protein reduces tumor burden.
Common side effects include fatigue, anemia, headache, and nausea. It may cause severe anemia requiring transfusions and can induce hypoxia. The medication carries embryo-fetal toxicity risk; effective contraception is required during treatment and for 1 month after the last dose. Patients should be monitored for new primary malignancies.
