Prevents bone complications (fractures, spinal compression, radiation/surgery to bone) in patients with solid tumors metastatic to bone; treats adults and skeletally mature adolescents with giant cell tumor of bone that is unresectable or where surgery would be harmful; treats hypercalcemia of malignancy refractory to bisphosphonates.

Denosumab binds to RANK ligand (RANKL), a protein essential for bone-removing cells (osteoclasts), preventing RANKL from activating its receptor and thereby reducing bone destruction.

Carries risk of hypocalcemia, especially in patients with kidney impairment; associated with osteonecrosis of the jaw (ONJ); not for patients with hypocalcemia; must be given by subcutaneous injection by a healthcare professional; patients should maintain adequate calcium and vitamin D intake.
