Yondelis is FDA-approved for treating advanced soft tissue sarcoma in patients who have previously received anthracycline-containing chemotherapy, and for treating relapsed platinum-sensitive ovarian cancer in combination with pegylated liposomal doxorubicin (PLD).
Trabectedin, the active ingredient in Yondelis, is an alkylating agent that binds to the minor groove of DNA, causing DNA damage that disrupts transcription and cell division in cancer cells, ultimately leading to cell death.
Yondelis is administered as a 24-hour intravenous infusion every 3 weeks and carries significant risks including severe liver toxicity, neutropenia, muscle damage (rhabdomyolysis), and heart failure. It should only be given under the supervision of healthcare professionals experienced in chemotherapy, and regular blood tests are required to monitor for side effects. The medication is contraindicated during pregnancy and breastfeeding.
